The application of conventional organic solvents has been essential in several steps of bioprocesses in order to achieve sufficient economic efficiency. The use of organic solvents is frequently used either to partly or fully replace water in the reaction medium or as a process aid for downstream separation.Nowadays, manufacturers are increasingly requested to avoid and substitute solvents with hazardous potential. Therefore, the solvent selection must account for potential environmental hazards, health and safety problems, in addition to fulfilling the ideal characteristics for application in a process.For the first time, criteria including Environment, Health and Safety (EHS), as well as the technical requirements for reaction and separation have been reviewed, collected and integrated in a single organic solvent screening strategy to be used as a guideline for narrowing down the list of solvents to test experimentally. Additionally, we have also included a solvent selection guide based on the methodology developed in the Innovative Medicines Initiative CHEM21 (IMI CHEM21) project and applied specifically to water-immiscible solvents commonly used in bioprocesses. 相似文献

A new neurological mouse mutation that arose spontaneously in a BALB/cByJ stock displays a semidominant pattern of inheritance.
In the heterozygote, this mutation results in an early loss of Purkinje cells in the cerebellum, which is followed by the
overt symptom of an ataxic gait first observed at postnatal day 13 (P13). A portion of animals homozygous for the mutation
die within P0; the remaining homozygotes die by P25. The mutation maps to mouse Chromosome (Chr) 6 between markers D6Rck314 and D6Rck361, a chromosomal segment that contains the lurcher (Lc) locus. The Lc mutation is also semidominant and has a strikingly similar phenotype. A cross between a new mutant (Nm) heterozygote and an Lc heterozygote yields double heterozygotes, animals that carry both mutations, with a phenotype similar to that of both Nm and Lc homozygotes. The similarity in phenotype, the colocalization of the two loci on mouse Chr 6, and the positive result of the
allelism test demonstrate that the new mutation is an allele of the Lc gene.

Replication of the Chinese hamster dihydrofolate (dhfr) gene initiates near a 281-bp HaeIII fragment of stably bent DNA that binds RIP60, a 60-kDa origin-specific DNA-binding protein that has been purified from HeLa cell nuclear extract (L. Dailey, M. S. Caddle, N. Heintz, and N. H. Heintz, Mol. Cell. Biol. 10:6225-6235, 1990). Circular permutation assays showed that stable DNA bending in the dhfr origin region fragment was due to the presence of five oligo (dA)3-4 tracts, designated bend elements B1 to B5, that are spaced 10 bp apart. DNA bending directed by elements B1 to B5, as assessed by anomolous migration of DNA fragments on polyacrylamide gels, was accentuated at 4 degrees C. Bend element B5, which is in inverse orientation relative to elements B1 to B4, overlaps an ATT-rich motif that comprises the RIP60 protein-binding site. Gel mobility shift assays with circularly permuted bent DNA fragments and purified RIP60 showed that RIP60 markedly enhanced DNA bending of the dhfr origin region sequences. These results suggest that, as in many plasmids, bacteriophages, and eucaryotic viruses, mammalian DNA-binding proteins may enhance DNA bending near origins of replication during initiation of DNA synthesis. 相似文献

Deoxycholate-KCl washed reticulocyte ribosomes are purified by affinity chromatography using Sepharose columns to which polyuridylic acid has been covalently bound. Upon passage through the column under nonenzymic conditions (high Mg2+:K+ ratio) approximately 10% of the ribosomes are retained. These ribosomes are then eluted with a buffer containing a high K+:Mg2+ ratio and are assayed for activity in various steps of the elongation process of protein synthesis. The lurcher (Lc) mutant mouse strain exhibits postnatal degeneration of cerebellar Purkinje cells. We have typed progeny from an intersubspecific, phenotypic backcross at seven loci to develop a genetic linkage map which spans approximately 35 cM surrounding and including the Lc locus on mouse chromosome 6. [(Mus musculus castaneus x B6CBA-Aw-J/A-Lc)F1 x B6CBA-Aw-J/A]N2 progeny were scored visually for the lurcher phenotype and molecularly, through restriction fragment length polymorphism analysis, for six cloned markers. Two candidate genes, Npy and Pcp-1, which map to mouse chromosome 6 and which are expressed in the cerebellum, are demonstrated to be distinct from Lc. Three genes are shown to be closely linked to the Lc locus, and the map order cen-Cpa-Npy-Cbl-1-Lc-Igk, Fabpl-Pcp-1 is determined.
